Description
- Herceptin 440 mg Injection is indicated for adjuvant treatment of HER2 overexpressing breast cancer, metastatic breast cancer, and metastatic gastric or gastroesophageal junction adenocarcinoma.
Dosage and administration
- Administer according to recommended doses and schedules for adjuvant or metastatic treatment. Reconstitute with diluent, administer intravenously, and store reconstituted solution in refrigerator.
Pharmacology
- Trastuzumab inhibits proliferation of human tumor cells that overexpress HER2 and mediates antibody-dependent cellular cytotoxicity (ADCC) preferentially on HER2 overexpressing cancer cells.
Interaction
- Increased risk of cardiac dysfunction if anthracycline is used after stopping Herceptin. Monitor cardiac function carefully if anthracyclines are used.
Side effects
- Serious adverse reactions include Cardiomyopathy, Infusion Reactions, Embryo-Fetal Toxicity, Pulmonary Toxicity, and exacerbation of Chemotherapy-Induced Neutropenia. Common adverse reactions include fever, nausea, vomiting, and fatigue.
Pregnancy and lactation
- Can cause fetal harm. Verify pregnancy status prior to initiation, advise effective contraception during treatment and for 7 months following the last dose. No information on presence in human milk or effects on breastfed infant.
Precautions and warnings
- Can cause Cardiomyopathy, conduct thorough cardiac monitoring. Infusion reactions may occur, monitor patients and consider permanent discontinuation in case of severe reactions. Can cause embryo-fetal toxicity and pulmonary toxicity. Exacerbation of Chemotherapy-Induced Neutropenia may occur.
Special populations
- Safety and effectiveness in pediatric patients have not been established.
Reconstitution
- Reconstitute with specified diluent and preservative, use immediately or store in refrigerator, discard unused portion after specified duration.
Storage conditions
- Store in original carton at specified temperature, protect from light, and keep out of the reach of children. Diluted solution should be used within specified duration and should not be frozen.
