Indications

Pharmacology

  • Proton pump inhibitor
  • Suppresses gastric acid secretion by specific inhibition of the H+/K+-ATPase
  • Provides better acid control than racemic proton pump inhibitors
  • Absorption occurs at approximately 1.5 hours after oral administration
  • Extensively metabolized in the liver by the cytochrome P450 (CYP) enzyme system
  • Plasma elimination half-life is approximately 1–1.5 hours

Dosage

  • Oral dosage form for various conditions
  • 20 mg or 40 mg Once Daily for 4-8 Weeks for healing of erosive esophagitis
  • 20 mg Once Daily for maintenance of healing of erosive esophagitis
  • 20 mg Once Daily for Symptomatic GERD
  • Triple Therapy dosage for Helicobacter Pylori eradication
  • Dose range of 20-80 mg for Zollinger-Ellison Syndrome

Administration

  • Tablet or capsule should be swallowed whole and taken one hour before a meal

Interaction

  • Extensively metabolized in the liver by CYP2C19 and CYP3A4

Contraindications

Pregnancy & Lactation

  • No adequate and well-controlled studies in pregnant women
  • Animal studies have revealed no teratogenic effects
  • Breast-feeding should be discontinued if the use of esomeprazole is considered essential

Precautions & Warnings

  • Symptomatic response to therapy with Esotron does not preclude the presence of gastric malignancy
  • Specific instructions for patient information and use with antacids

Use in Special Populations

  • No established safety and effectiveness in pediatric patients
  • No overall differences in safety and efficacy observed between the elderly and younger individuals
  • No dosage adjustment recommended for patients with mild to moderate hepatic insufficiency
