Indications
- Osteoarthritis (OA)
- Rheumatoid arthritis (RA)

- Moderate pain associated with dental surgery
Pharmacology
- Orally active cyclooxygenase-2 (COX-2) specific inhibitor
- Inhibition of COX-2 decreases pain and inflammation
- Selective inhibition of COX-2 decreases potential for GI toxicity
Dosage & Administration
- Recommended doses for osteoarthritis, rheumatoid arthritis, ankylosing spondylitis, acute gouty arthritis, and postoperative dental surgery pain
- Shortest duration and lowest effective daily dose should be used
- Need for symptomatic relief and response to therapy should be re-evaluated periodically
Interactions
- Increase in prothrombin time with oral anticoagulants
- Possible reduction in the effect of diuretics and other antihypertensive drugs
- Concomitant use with low-dose acetylsalicylic acid for cardiovascular prophylaxis
- Potential increase in nephrotoxic effect of ciclosporin or tacrolimus
- Decreased lithium renal excretion leading to increased lithium plasma levels
Contraindications
- Hypersensitivity to the active substance or excipients
- Active peptic ulceration or active GI bleeding
- History of allergic-type reactions after taking acetylsalicylic acid or NSAIDs
- Pregnancy, lactation, severe hepatic dysfunction, estimated renal creatinine clearance <30 ml/min, children and adolescents under 16 years of age, inflammatory bowel disease, congestive heart failure, uncontrolled hypertension, established ischaemic heart disease, peripheral arterial disease, and/or cerebrovascular disease

Pregnancy & Lactation
- Use not recommended in women attempting to conceive
- Excretion in the milk of lactating rats
- Breastfeeding is not recommended
Precautions & Warnings
- Caution in treating patients at risk of developing GI complications with NSAIDs
- Only treat patients with significant cardiovascular risk factors after careful consideration
- Monitoring of renal function in patients with decreased prostaglandin formation and impaired renal blood flow
- Exercise caution in patients with a history of cardiac failure, left ventricular dysfunction, hypertension, pre-existing edema
- Monitoring of liver function and discontinuation if signs of hepatic insufficiency occur
- Discontinue at the first appearance of hypersensitivity
- Caution when co-administering with warfarin or other oral anticoagulants
Overdose Effects
- No significant toxicity observed with single or multiple doses
- Usual supportive measures recommended in case of overdose
