Indications

Pharmacology
Naproxen blocks COX-1 and COX-2 enzymes leading to decreased prostaglandin synthesis. COX-2 enzyme mediates antipyretic, analgesic, and anti-inflammatory properties, while undesired adverse effects are linked to COX-1 enzyme.
Dosage & Administration
- Naproxen oral preparations should be taken with water after meals.
- Dosage varies for different indications and age groups.

Contraindications
Contraindicated in patients with known hypersensitivity to Naproxen. Avoid in patients with asthma or allergic-type reactions after taking aspirin or other NSAIDs. Contraindicated for perioperative pain in coronary artery bypass graft (CABG) surgery.
Side Effects
- Gastrointestinal: Heartburn, abdominal pain, nausea, diarrhea, dyspepsia
- Central Nervous System: Headache, vertigo, drowsiness
- Dermatological: Pruritus (itching), purpura
- Cardiovascular: Edema, palpitation
- Others: Visual disturbances, hearing disturbances
Pregnancy & Lactation
Naproxen may have adverse effects on the fetus. Use in first and third trimesters requires careful balancing of benefits to mother against possible risks to the fetus. Excreted in small amounts in breast milk.
Precautions & Warnings
- Renal effects: Use with caution in patients with impaired renal function. Monitor serum creatinine and/or clearance. Caution with creatinine clearance of less than 20ml/min.
- Hepatic effects: Reports of moderate to severe jaundice attributed to Naproxen.
