Medicine Details

Indications

  • Bronchial asthma
  • Bronchospasm
  • Chronic obstructive pulmonary disease (COPD)
  • Pulmonary disease with spastic bronchial component

Pharmacology

  • Doxophylline is a novel bronchodilator
  • Selectively inhibits phosphodiesterase 4
  • Relaxes bronchial smooth muscle
  • Reported to inhibit platelet activating factor (PAF) and generation of leukotriene production

Dosage & Administration

  • Elderly: 200 mg tablet two or three times daily
  • Adults: 400 mg tablet two or three times daily or as prescribed by the physician
  • Children >12 years of age: 10 ml syrup or 200 mg tablet two or three times daily
  • Children 6-12 years of age: 6-9 mg/kg body weight two times daily
  • If required daily dose of Doxophylline is 400 mg then Doxophylline SR tablet to be taken once daily

Interaction

  • Should not be administered with other xanthine preparations
  • Limit consumption of beverages and food containing caffeine
  • Caution with ephedrine or other sympathomimetic drugs
  • Concurrent administration with erythromycin, TAO, lincomycin, clindamycin, allopurinol, cimetidine, influenza vaccine, propanolol
  • Phenytoin, other anticonvulsants, and cigarette smoking may increase the clearance of xanthine derivatives

Contraindications

  • Known hypersensitivity to the drug or other xanthine derivatives
  • Acute myocardial infarction
  • Hypotension
  • During lactation

Pregnancy & Lactation

  • Use should be evaluated carefully during pregnancy
  • Contraindicated during lactation
  • Does not affect the ability to drive or use machines

Precautions & Warnings

  • Factors reducing hepatic clearance
  • Age, congestive cardiac decompensation, chronic obstructive pulmonary disease, severe liver disease, concomitant infections
  • Caution in patients with cardiac disease, hypertension, elderly, severe hypoxemia, hyperthyroidism, chronic corpulmonale, congestive heart failure, liver disease, peptic ulcer, renal impairment
  • Monitoring of blood levels recommended
  • No risk of addiction or dependence

Overdose Effects

  • Symptomatic treatment of cardiovascular collapse should be instituted
  • No specific antidote
