Description

Indications

  • Relieve chronic heartburn symptoms
  • Healing of erosive esophagitis
  • Maintenance of healing of erosive esophagitis
  • Eradication of Helicobacter pylori infection
  • Treatment of Zollinger-Ellison Syndrome
  • Acid-related Dyspepsia
  • Treatment of duodenal and gastric ulcers

Pharmacology

  • Proton pump inhibitor
  • Suppression of gastric acid secretion
  • Better acid control than racemic proton pump inhibitors
  • Enteric-coated pellet formulation
  • Peak plasma levels at approximately 1.5 hours after oral administration
  • Extensive metabolism in the liver
  • 97% bound to plasma proteins
  • Approximately 1-1.5 hours plasma elimination half-life
  • Excretion of less than 1% in the urine

Dosage

  • 20 mg or 40 mg once daily for healing of erosive esophagitis
  • 20 mg once daily for maintenance of healing of erosive esophagitis
  • 20 mg once daily for symptomatic GERD
  • Triple therapy for Helicobacter pylori eradication
  • 20-80 mg once daily for Zollinger-Ellison Syndrome
  • 20-40 mg once daily for acid-related dyspepsia
  • 20 mg once daily for 2-4 weeks for duodenal ulcer
  • 20-40 mg once daily for 4-8 weeks for gastric ulcer
  • IV injection or infusion for various conditions based on patient age and weight

Administration

  • Tablet or capsule should be swallowed whole and taken one hour before a meal
  • Oral suspension instructions
  • IV injection preparation and administration

Interaction

  • Extensively metabolized in the liver by CYP2C19 and CYP3A4
  • Unlikely to inhibit certain CYP enzymes
  • Potential interference with drugs influenced by gastric pH

Contraindications
