Pharmacology
- Proton pump inhibitor
- Suppression of gastric acid secretion
- Specific inhibition of the H+/K+-ATPase
- Better acid control than racemic proton pump inhibitors
- Absorption: peak plasma levels at approximately 1.5 hours
- Distribution: 97% bound to plasma proteins
- Metabolism: extensive metabolism in the liver
- Excretion: plasma elimination half-life of approximately 1–1.5 hours
Dosage
- Oral dosage form for various conditions
- IV Injection or IV infusion for different patient groups
Administration
- Tablet or capsule should be swallowed whole and taken one hour before a meal

Interaction
- Extensive metabolism by CYP2C19 and CYP3A4
- No clinically relevant interactions with certain drugs
- Potential interference with CYP2C19 and gastric pH dependent drugs
- Specific interactions with oral contraceptives, diazepam, phenytoin, or quinidine
- Increased plasma levels of Esoflux and 14-hydroxyclarithromycin
Contraindications
- Known hypersensitivity to any of the formulation
Side Effects
- Frequently occurring adverse events: headache, diarrhea, nausea, flatulence, abdominal pain, constipation, dry mouth
- No difference in types of related adverse events seen during maintenance treatment
Pregnancy & Lactation
- No adequate and well-controlled studies in pregnant women
- Animal studies revealing no teratogenic effects
- Excretion of esomeprazole in milk not studied
- Discontinuation of breastfeeding if esomeprazole use is considered essential
Precautions & Warnings
- Symptomatic response does not preclude the presence of gastric malignancy
- Specific instructions for patients with difficulty swallowing capsules
- Allowed use of antacids while taking Esoflux
Use in Special Populations
- Safety and effectiveness in paediatric patients not established
- No overall differences in safety and efficacy in the elderly
- No dosage adjustment recommended for patients with mild to moderate hepatic insufficiency
- Pharmacokinetics not expected to be altered in patients with renal impairment
Overdose Effects
- Acute toxic signs include reduced motor activity, respiratory changes, tremor, ataxia, clonic convulsions
- No specific antidote known, symptomatic and supportive treatment recommended

- Temperature not exceeding 30°C

- Keep out of reach of children
