Description
- Avastrol is indicated as an adjunct to diet to reduce elevated total cholesterol, LDL cholesterol, apolipoprotein B (Apo-B) and triglycerides levels in various conditions.
Function
- Reduces elevated total cholesterol
- Lowers LDL cholesterol and apolipoprotein B levels
- Reduces triglycerides levels
- Prevents cardiac ischaemic events
- Adjunct to diet for various dyslipidemias
Dosage & Administration
- 10 mg or 20 mg once daily
- Dosage may be adjusted according to patient response
- Administration with or without food
- Dose adjustment based on lipid levels within 2 to 4 weeks
Pharmacology
- Selective inhibitor of HMG-CoA reductase
- Lowers plasma cholesterol and lipoprotein levels
- Increases hepatic LDL receptors for enhanced uptake and catabolism of LDL
Absorption
- Rapid absorption after oral administration
- Maximum plasma concentrations occur within 1 to 2 hours
Distribution
- Mean volume of distribution approximately 381 liters
- 98% bound to plasma proteins
Metabolism
- Extensively metabolized to ortho- and parahydroxylated derivatives and beta-oxidation products
- Metabolism by cytochrome P450 3A4
- Active metabolites contribute to circulating inhibitory activity for HMG-CoA reductase
Excretion
- Eliminated primarily in bile following hepatic and/or extra-hepatic metabolism
- Mean plasma elimination half-life approximately 14 hours
Interaction
- Increased risk of myopathy with concurrent administration of specific drugs
- Plasma concentration alterations with coadministration of antacid, colestipol, digoxin, erythromycin, oral contraceptives, warfarin
Contraindications
- Hypersensitivity to any component of the medication
- Active liver disease or unexplained persistent elevations of serum transaminases
- History of serious adverse reaction to prior administration of HMG-CoA reductase inhibitors
