Pharmacology
Triazole antifungal agent, inhibitor of fungal cytochrome P-450 dependent enzymes
Dosage & Administration
- Adults:
- Specific dosages for various indications
- Dosage modifications based on age and renal impairment
- Child over 1 year:
- Dosages for different indications
- Dosage based on weight
- Elderly patient: Normal dose usage if no evidence of renal impairment
- Renal Impairment: No adjustment in single dose therapy, dosage modification in multiple dose therapy based on creatinine clearance and dialysis
Interaction
- Effect on prothrombin time after warfarin administration
- Prolongation of serum half-life of concomitantly administed oral sulphonyl ureas
- Increase in plasma concentrations of fuconazole by hydrochlorothiazide
- Increase in phenytoin levels
- Effect on combined oral contraceptive, cyclosporin, theophyline, and other medications

Hypersensitivity to Fluconazole or related triazole compounds

Pregnancy & Lactation
Adverse fetal effects seen in animals at dose levels associated with maternal toxicity
Precautions & Warnings
- Abnormalities of hepatic, renal, haematological and other biochemical function tests in some patients
- Rare occurrences of hepatic necrosis
- Exfoliative cutaneous reactions
- Risk-benefit ratio assessment in patients with significant rise of liver enzymes
- Use during lactation not recommended
- Unlikely to impair ability to drive or use machinery

Overdose Effects
- Supportive measures and symptomatic treatment
- Gastric lavage if necessary
- Forced volume diuresis to increase elimination rate
- Haemodialysis to decrease plasma levels
