Description
- Selective COX-2 Inhibitor
- Symptomatic relief for Osteoarthritis, Rheumatoid arthritis, Ankylosing spondylitis, and acute gouty arthritis
- Short-term treatment for moderate pain associated with dental surgery
- Orally active cyclooxygenase-2 (COX-2) specific inhibitor
- Potent and orally active
- Decreases clinical signs and symptoms with decreased potential for GI toxicity and effects on platelet aggregation
- Dosage forms: 30 mg, 60 mg, 90 mg, 120 mg

Indications
- Relief of pain and signs of inflammation associated with various conditions like OA, RA, ankylosing spondylitis, and acute gouty arthritis
- Short-term treatment of moderate pain associated with dental surgery
Pharmacology
- Inhibition of COX-2 without inhibition of COX-1
- Decreased potential for GI toxicity and effects on platelet aggregation
Dosage and administration
- Recommended doses for different conditions: 30 mg once daily, 60 mg once daily, 90 mg once daily, 120 mg once daily
- Duration of treatment for acute gouty arthritis: 8 days
- Limited to a maximum of 3 days for postoperative dental surgery pain
Interaction
- Potential interactions with oral anticoagulants, diuretics, ACE inhibitors, Angiotensin II Antagonists, Acetylsalicylic Acid, Ciclosporin, Tacrolimus, and Lithium
Contraindications
- Hypersensitivity to the active substance or excipients
- Active peptic ulceration or active GI bleeding
- Patients with specific allergic-type reactions after taking acetylsalicylic acid or NSAIDs
- Pregnancy, lactation
- Severe hepatic dysfunction, estimated renal creatinine clearance <30 ml/min
- Children and adolescents under 16 years of age
- Inflammatory bowel disease
- Congestive heart failure, hypertension, established ischaemic heart disease, peripheral arterial disease, and/or cerebrovascular disease

Precautions and warnings
- Caution advised for patients at risk of developing GI complications with NSAIDs
- Special consideration for patients with significant risk factors for cardiovascular events
- Caution for patients with renal impairment, hepatic dysfunction, cardiac failure, left ventricular dysfunction, or hypertension
- Regular monitoring of renal and liver function
- Discontinue at the first appearance of skin rash, mucosal lesions, or hypersensitivity
- May mask fever and other signs of inflammation
- Caution when co-administering with warfarin or oral anticoagulants
Overdose effects
- Single doses up to 500 mg and multiple doses up to 150 mg/day did not result in significant toxicity
- Usual supportive measures recommended in the event of overdose
