Indications
- Treats bronchial asthma

Pharmacology
- Novel bronchodilator
- Structurally differs from Theophylline
- Selective inhibition of phosphodiesterase 4
- Relaxes bronchial smooth muscle
- Decreased affinities toward adenosine A1 and A2 receptors
- Inhibits platelet activating factor (PAF) and generation of leukotriene production
Dosage & Administration
- Elderly dosage: 200 mg tablet two or three times daily
- Adult dosage: 400 mg tablet two or three times daily
- Children dosage: varies based on age and body weight
- Doxophylline SR tablet for required daily dose of 400 mg
Interaction
- Should not be administered with other xanthine preparations
- Limit consumption of caffeine-containing beverages and food
- Caution in administering with ephedrine or other sympathomimetic drugs
- Concurrent administration with erythromycin, TAO, lincomycin, clindamycin, allopurinol, cimetidine, influenza vaccine, propanolol and their effects on hepatic clearance
- Effects of phenytoin, other anticonvulsants, and cigarette smoking on clearance of xanthine derivatives
Contraindications
- Known hypersensitivity to the drug or other xanthine derivatives
- Acute myocardial infarction
- Hypotension
- During lactation

- Extrasystoles
- Tachypnea
- Hyperglycemia
- Albuminuria
- Possible severe cardiac arrhythmias and tonic-clonic seizure in case of overdose
Pregnancy & Lactation
- Animal tests show no interference with pre and postnatal growth
- Use during pregnancy requires careful evaluation of risk-benefit ratio
- Contraindicated during lactation
- No effects on ability to drive and use machines
Precautions & Warnings
- Factors affecting hepatic clearance of xanthine derivatives
- Caution in patients with cardiac disease, hypertension, hypoxemia, hyperthyroidism, corpulmonale, congestive heart failure, liver disease, peptic ulcer, and renal impairment
- Special caution in patients with congestive heart failure
- No risk of addiction or dependence
Overdose Effects
- Possible severe cardiac arrythmias and tonic-clonic seizure
- No specific antidote
