Indications
- spasticity resulting from multiple sclerosis
- flexor spasms and concomitant pain
- skeletal muscle spasm resulting from rheumatic disorders
- spinal cord injuries
- cerebrovascular accidents
- neoplastic or degenerative brain disease
Pharmacology
- inhibits monosynaptic and polysynaptic reflexes
- stimulates GABA receptors
- inhibits release of glutamate and aspartate
- produces CNS depression
- antinociceptive effect
Dosage & Administration
- 5 mg 3 times daily for adults & children over 10 years
- gradually increased dosage
- max. 100 mg daily for adults & children over 10 years
- treatment usually started with 2.5 mg for children <10 years
- dosage raised according to requirement for children <10 years
- maintenance dose based on age
Interaction
- increased sedation with agents acting on the central nervous system
- increased sedation with alcohol or synthetic opiates
- increased risk of respiratory depression
- fall in blood pressure with antihypertensives
- potentiated effects with tricyclic antidepressants
- potential side effects when combined with levodopa and carbidopa
- increased CNS depressant effects with MAO inhibitors
- synergistic effect with magnesium sulphate or neuromuscular blocking agents
Contraindications
- hypersensitivity to any component

- adverse reactions in various systems
Pregnancy & Lactation
- Pregnancy category B3
- safe use during pregnancy not established
- crosses the placental barrier
- should only be administered when benefits outweigh possible hazards
- excreted in breast milk
- quantities in breast milk expected to have no undesirable effects on the infant
Precautions & Warnings
- lower doses for patients with impaired renal function or undergoing chronic haemodialysis
- closely monitor patients with psychotic disorders, depressive or manic disorders
- caution in patients with epilepsy
- care with patients with peptic ulcers, cerebrovascular diseases, or hepatic, renal, or respiratory failure
- monitor respiratory and cardiovascular function
- caution in patients with preexisting sphincter hypertonia
- poor tolerance in patients with stroke
- appropriate laboratory tests for patients with hepatic diseases or diabetes mellitus
Overdose Effects
- gastric lavage in case of severe overdose
