Indications
- Reduce elevated total cholesterol levels
- Reduce LDL cholesterol levels
- Reduce apolipoprotein B levels
- Reduce triglycerides levels
- Treat heterozygous familial hypercholesterolaemia
- Treat homozygous familial hypercholesterolaemia
- Treat mixed dyslipidemia
- Treat hypertriglyceridaemia
- Treat dysbetalipoproteinaemia
- Reduce cardiac ischaemic events
- Treatment of hypercholesterolemia associated with diabetes mellitus
- Treatment of hypercholesterolemia associated with renal transplantation
Pharmacology
- Selective inhibitor of HMG-CoA reductase
- Lowers plasma cholesterol and lipoprotein levels
- Inhibits cholesterol synthesis in the liver
- Increases hepatic LDL receptors for enhanced uptake and catabolism of LDL
Dosage & Administration
- Adult dosage of 10-80 mg once daily
- Child dosage of 10-20 mg once daily
- Starting dose of 10 or 20 mg once daily
- Dosage adjustment based on lipid levels analysis within 2 to 4 weeks
- Pediatric dosage of 10-20 mg/day
- Individualized dosing for pediatric patients
- Administration with or without food
Interaction
- Increased risk of myopathy with concurrent administration of certain drugs
- Variation in plasma concentrations when coadministered with antacids, colestipol, digoxin, erythromycin, oral contraceptives, and warfarin
Contraindications
- Hypersensitivity to any component of the medication
- Active liver disease or unexplained persistent elevations of serum transaminases
- History of serious adverse reaction to HMG-CoA reductase inhibitors

Pregnancy & Lactation
- Atorvastatin is contraindicated during pregnancy

Precautions & Warnings
- Liver function tests before initiation of treatment and periodically thereafter
- Discontinue therapy if markedly elevated CPK levels occur or myopathy is diagnosed or suspected
Use in Special Populations
- Higher plasma concentrations in elderly subjects
- No clinically significant difference in LDL-C reduction between men and women
- No influence of renal disease on plasma concentrations or LDL-C reduction
- Markedly increased plasma concentrations in patients with chronic alcoholic liver disease

- Symptomatic treatment and supportive measures instituted
- Liver function tests and serum CK levels monitoring recommended
