Description
- A medication indicated for relief from chronic heartburn
- Used for healing erosive esophagitis and for its maintenance
- Eradicates Helicobacter pylori infection in patients with duodenal ulcer disease
- Effective in treating Zollinger-Ellison Syndrome, acid-related dyspepsia, duodenal and gastric ulcers

PharmacologyMechanism
- Proton pump inhibitor that suppresses gastric acid secretion
- Inhibits the H+/K+-ATPase in the gastric parietal cell
- Es-isomer providing better acid control
- Absorption occurs at approximately 1.5 hours (Tmax)
- Metabolized in the liver by the CYP enzyme system
AdministrationInstructions
- Swallow whole and take one hour before a meal for tablet or capsule
- Mix contents of the oral suspension with water
- IV injection prepared with 0.9% Sodium Chloride and administrated intravenously over at least 3 minutes
Interaction
- Extensively metabolized in the liver by CYP2C19 and CYP3A4
- Not likely to inhibit CYPs 1A2, 2A6, 2C9, 2D6, 2E1 and 3A4
- Interferes with the absorption of certain drugs
- No clinically significant interactions with phenytoin, warfarin, quinidine, clarithromycin or amoxicillin
Contraindications
- Contraindicated in patients with known hypersensitivity to any of the formulation

- Lethal dose to rats observed at 103 times the human dose on a body surface area basis
- Signs of acute toxicity include reduced motor activity, changes in respiratory frequency, tremor, ataxia, and intermittent clonic convulsions
- No reports of overdose with Esigerd
- No specific antidote known
