Indications

Description
- Selective inhibitor of cyclic guanosine monophosphate (cGMP)-specific phosphodiesterase type 5 (PDE5)
- Requires sexual stimulation to be effective
Mechanism of Action
- Local release of nitric oxide
- Increased levels of cGMP in corpus cavernosum
- Smooth muscle relaxation
- Inflow of blood into penile tissues
Pharmacokinetics
- Rapid absorption after oral administration
- Mean absolute bioavailability of 41%
- Eliminated predominantly by hepatic metabolism
- Converted to active metabolite with properties similar to the parent drug
- Terminal half lives of about 4 hours
Dosage & Administration
- Recommended dose of 50 mg taken 1 hour before sexual activity
- Dose may be increased to a maximum of 100 mg or decreased to 25 mg
- Maximum recommended dosing frequency is once per day
- Special dosing considerations for specific patient groups
Metabolism & Interaction
- Principally mediated by cytochrome P450 isoforms 3A4 and 2C9
