Description
- A proton pump inhibitor for relieving symptoms associated with chronic heartburn, GERD, erosive esophagitis, and other gastric conditions
- Used in combination with antibiotics for Helicobacter pylori eradication and for the management of Zollinger-Ellison Syndrome, acid-related dyspepsia, duodenal and gastric ulcers
Indications
- Relief from chronic heartburn symptoms
- Healing of erosive esophagitis
- Maintenance of healing of erosive esophagitis
- Eradication of Helicobacter pylori infection
- Treatment of Zollinger-Ellison Syndrome
- Management of acid-related dyspepsia
- Treatment of duodenal and gastric ulcers
Pharmacology
- DrugAction:
- Suppression of gastric acid secretion
- Absorption:
- Peak plasma levels occur at approximately 1.5 hours after oral administration
- Systemic bioavailability is approximately 90% at steady state
- Plasma protein binding is constant over a specified concentration range
- Metabolism:
- Extensively metabolized in the liver by the cytochrome P450 enzyme system
- Excretion:
- Plasma elimination half-life is approximately 1–1.5 hours
- Less than 1% of the parent drug is excreted in the urine
Dosage
- Oral:
- Dosage forms: 20 mg or 40 mg capsules
- Healing of erosive esophagitis: 20 mg or 40 mg once daily for 4-8 weeks
- Maintenance of healing of erosive esophagitis: 20 mg once daily
- Symptomatic GERD: 20 mg once daily for 4 weeks
- Iv:
- Dosage forms: Injection or infusion
- Dosage for adult patients (≥18 years): 20 mg or 40 mg once daily
- Dosage for pediatric patients (1 month to <1 year): 0.5 mg/kg once daily
Administration
- TabletOrCapsule: Swallowed whole and taken one hour before a meal
- OralSuspension: Should be prepared and consumed within a specified timeframe
- IvInjection: Administered intravenously over a period of at least 3 minutes
