Description
- Anzitor is indicated as an adjunct to diet to reduce elevated total cholesterol, LDL cholesterol, apolipoprotein B (Apo-B) and triglycerides levels.
Dosage
- 10 mg once daily for adults, 10-80 mg once daily for adults with primary hypercholesterolaemia and combined hyperlipidaemia.
Pharmacology
- Atorvastatin is a selective inhibitor of HMG-CoA reductase, lowering plasma cholesterol and lipoprotein levels.
Absorption
- Rapidly absorbed after oral administration; maximum plasma concentrations occur within 1 to 2 hours.
Distribution
- Mean volume of distribution of Atorvastatin is approximately 381 liters.
Metabolism
- Extensively metabolized to ortho- and parahydroxylated derivatives and various beta-oxidation products.
Excretion
- Atorvastatin and its metabolites are eliminated primarily in bile following hepatic and/or extra-hepatic metabolism.
Interactions
- Risk of myopathy increased with concurrent administration of cyclosporine, fibric acid derivatives, niacin, erythromycin, azole antifungals.
Contraindications
- Hypersensitivity to any component, active liver disease, unexplained persistent elevations of serum transaminases, serious adverse reaction to prior administration of HMG-CoA reductase inhibitors.
Side Effects
- Constipation, flatulence, dyspepsia, abdominal pain, infection, headache, back pain, rash, asthenia, arthralgia, myalgia.
Pregnancy & Lactation
- Atorvastatin is contraindicated during pregnancy and lactation.
Precautions & Warnings
- Liver function tests should be performed before initiation of treatment; Anzitor should be used with caution in patients consuming substantial quantities of alcohol or with a history of liver disease.
Use in Special Populations
- Plasma concentrations of Anzitor are higher in healthy elderly subjects; no clinically significant difference in LDL-C reduction between men and women.
Overdose Effects
- Specific treatment not available; liver function tests should be performed and serum CK levels monitored.
