Indications

Composition
- Thiopental 500 mg injection
- Thiopental 1 gm injection
- Sterile mixture of Thiopental Sodium and anhydrous Sodium Carbonate

Pharmacology
- Ultrashort-acting depressant of the central nervous system
- Induces hypnosis and anesthesia
- Binds at a distinct binding site associated with a Cl- ionopore at the GABAA receptor
- Increases duration of time for which the Cl- ionopore is open
- Prolongs the post-synaptic inhibitory effect of GABA in the thalamus
Dosage & Administration
- Administered intravenously as a 2.5% w/v solution
- Can be administered as a 5% w/v solution
- Premedication with atropine or scopolamine
- Test dose for tolerance assessment
- Normal dosage for induction of anesthesia
- Dosage adjustments based on patient's response
- Specific dosages for adults, children, and elderly
- Dosage for convulsive states
Interaction
- Increase in difficulty in producing anesthesia with alcohol or CNS depressants
- Additive action with other CNS depressants
- Increased hypotension and excitatory effects with phenothiazine antipsychotics
- Increased hypnotic effect with antipsychotic
- Decreased requirement of TPS with specific drugs
Contraindications
- Absence of suitable veins for intravenous administration
- Hypersensitivity to barbiturates
- Variegate porphyria or acute intermittent porphyria
- Status asthmaticus
- Severe cardiovascular disease
- Hypotension or shock
- Conditions prolonging or potentiating the hypnotic effect
