Pharmacology
- Active against Gram-positive organisms
- Little activity against Gram-negative bacilli
- Inhibits formation of bacterial cell wall
- Isoxazolyl penicillin properties
- Resistance to hydrolysis by penicillinase
- Bactericidal antibiotic
- Interferes with peptidoglycan synthesis
- Weakens bacterial cell wall leading to rupture
Dosage
- Oral administration for adults: 250 mg four times daily
- Dosage may be doubled in severe infections
- Up to 8 gm daily for osteomyelitis and endocarditis
- Dosage for children: 1/2 of adult dose (2-10 years), 1/4 of adult dose (Under 2 years)
- Parenteral administration for adults: 250 mg-1 g four times daily
- All systemic doses may be doubled in severe infections
Administration
Oral doses should be administered 1 hour before meal
Interaction
Concurrent use of Fluking may result in increased level of Fluking in blood for prolonged period
Contraindications
Contraindicated in penicillin hypersensitive patients

- Gastrointestinal side effects (nausea, vomiting, diarrhoea, dyspepsia)
- Rashes
- Urticaria
- Purpura
- Fever
- Interstitial nephritis
- Hepatitis
- Cholestatic jaundice
Pregnancy & Lactation
- US FDA Pregnancy Category: B
- No adequate and well-controlled studies in pregnant women
- Drug excreted in human milk
- Caution during lactation
Precautions & Warnings
- Use with caution in patients with hepatic dysfunction
- Caution in patients with allergic diathesis
Use in Special Populations
Consider dose reduction in severe renal failure (creatinine clearance <10 ml/min)
