Description
- Erlotinib reversibly inhibits the kinase activity of Epidermal Growth Factor Receptor (EGFR)
- Chemical name: N-(3-ethynylphenyl)-6,7-bis(2-methoxyethoxy)4-quinazolinamine
DosageAndAdministration
- Recommended daily dose of Erlotinib for NSCLC: 150 mg on an empty stomach
- Recommended daily dose of Erlotinib for pancreatic cancer: 100 mg in combination with gemcitabine on an empty stomach
- Dosage reduction and increment guidelines based on drug interactions and concurrent cigarette smoking
Indications
- First-line treatment of metastatic Non-Small Cell Lung cancer (NSCLC) with EGFR exon 19 deletions or exon 21 substitution mutations
- Maintenance treatment of locally advanced or metastatic NSCLC
- Treatment of locally advanced or metastatic NSCLC after failure of prior chemotherapy regimen
- First-line treatment of locally advanced, unresectable, or metastatic pancreatic cancer in combination with gemcitabine
Interaction
- Interactions with anticoagulants, CYP3A4 inhibitors, CYP3A4 inducers, and drugs affecting gastric pH
SideEffects
- Common adverse reactions: rash, diarrhea, anorexia, fatigue, dyspnea, cough, nausea, vomiting
- Serious adverse reactions include ILD, renal failure, hepatotoxicity, gastrointestinal perforation, bullous and exfoliative skin disorders, myocardial infarction, cerebrovascular accident, and others
