Indications

  • Short-term management of moderate to severe acute post-operative pain
  • Relief of pain, inflammation, and light sensitivity after ocular surgery
  • Decrease in eye inflammation or redness caused by seasonal allergies

Description

  • Belongs to the pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
  • Inhibits prostaglandin synthesis as a peripherally acting analgesic
  • Pharmacokinetic property is linear
  • Highly protein bound and largely metabolized in the liver
  • Excretion occurs in the urine

Pharmacology

  • Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
  • Acts by inhibiting the cyclooxygenase enzyme system and hence inhibits prostaglandin synthesis
  • Demonstrates minimal anti-inflammatory effect at analgesic dose

Dosage & Administration

  • Tablet recommended dose is 10 mg every 4-6 hours for short-term use up to 7 days
  • Injection for moderately severe, acute pain
  • Single or multiple doses, on a regular or as-needed schedule

Interaction

  • Other NSAIDs or aspirin increase side effects
  • Anti-coagulants enhance anti-coagulant effect
  • Beta Blockers reduce anti-hypertensive effect
  • ACE Inhibitors increase risk of renal impairment
  • Methotrexate enhances toxicity of methotrexate

Contraindications

  • Hypersensitivity to Ketorolac or other NSAIDs
  • Children under 16 years of age
  • Prophylactic analgesic use before surgery

  • Specific caution for use of eye drops in patients with corneal epithelial breakdown and wearing contact lenses
