Indications

Pharmacology

  • Pioglitazone decreases insulin resistance
  • Metformin improves glucose tolerance

Dosage & Administration

  • Individualized antihyperglycemic therapy
  • Maximum recommended daily dose of Pioglitazone 45 mg and Metformin 2550 mg
  • Divided daily doses with meals

Interaction

  • May impair vit B12 absorption
  • Increased plasma levels with strong CYP2C8 inhibitors
  • Decreased plasma levels with CYP2C8 inducers
  • Reduced metabolism with cationic drugs

Contraindications

  • Renal disease or dysfunction
  • Known hypersensitivity to Pioglitazone or Metformin
  • Acute or chronic metabolic acidosis
