Indications
- Relief from chronic heartburn symptoms
- Healing of erosive esophagitis
- Maintenance of healing of erosive esophagitis
- Eradication of Helicobacter pylori infection

Pharmacology
- Proton pump inhibitor
- Suppression of gastric acid secretion
- Enteric-coated pellet formulation
- Specific inhibition of H+/K+-ATPase
- Better acid control than racemic proton pump inhibitors
- Absorption peak at approximately 1.5 hours
- Metabolized in the liver by CYP enzyme system
Dosage
- Multiple dosage forms - oral, IV injection, IV infusion
- Different dosages for different indications
- Oral administration should be at least one hour before meals
- Duration of treatment for each indication
Administration
- Tablet or capsule should be swallowed whole
- Oral suspension should be taken into a small glass containing water
- IV injection solution preparation and administration
Interaction
- Metabolized by CYP2C19 and CYP3A4
- Inhibition of gastric acid secretion
- Potential interference with drug absorption due to gastric pH
- No clinically significant interactions with certain drugs
Contraindications
- Contraindicated in patients with known hypersensitivity to any of the formulation

- No dosage adjustment recommended for patients with mild to moderate hepatic insufficiency
- No alteration in pharmacokinetics in renal impairment
Overdose effects
- Lethality in rats at high doses
- Signs of acute toxicity
- No specific antidote known
- Symptomatic and supportive treatment recommended
