Indications

  • Management of post-operative pain
  • Relieve pain, inflammation, and light sensitivity after ocular surgery
  • Decrease eye inflammation or redness caused by seasonal allergies

Description

  • Pyrrolo-pyrrole group drug
  • Non-steroidal anti-inflammatory drug (NSAID)
  • Chemical composition: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2-amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
  • Inhibits synthesis of prostaglandins
  • Peripherally acting analgesic
  • Pharmacokinetic property: linear
  • Highly protein bound
  • Mainly metabolized in the liver
  • Metabolites and unchanged drug excreted in urine

Pharmacology

  • Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
  • Inhibits cyclooxygenase enzyme system
  • Minimal anti-inflammatory effect at analgesic dose

Dosage & Administration

  • Recommended tablet dose: 10 mg every 4-6 hours for short-term use up to 7 days
  • Injection dosage for different age groups and conditions
  • Onset of analgesic effect within 30 minutes for IV or IM administration

Interaction

  • Increased side effects with other NSAIDs or aspirin
  • Enhanced anti-coagulant effect with anti-coagulants
  • Reduced anti-hypertensive effect with beta blockers
  • Increased risk of renal impairment with ACE inhibitors
  • Enhanced toxicity of methotrexate

Contraindications

  • Hypersensitivity to Ketorolac or other NSAIDs
  • Not for use in children under 16 years of age
  • Contradicted as a prophylactic analgesic before surgery

  • Discontinue use of topical NSAIDs in evidence of corneal epithelial breakdown
