Indications
- Neuropathic pain associated with diabetic peripheral neuropathy (DPN)
- Postherpetic neuralgia (PHN)
- Adjunctive therapy for the treatment of partial-onset seizures
- Fibromyalgia
- Neuropathic pain associated with spinal cord injury
Pharmacology
- Structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA)
- Binds with high affinity to the alpha2-delta site
- Anti-nociceptive and antiseizure effects
Dosage
- Maximum recommended dose of 300 mg/day
- Dosing based on efficacy and tolerability
- Starting dose of 50 mg three times a day
- Maximum recommended dose of 660 mg once daily
Administration
- Oral route of administration
- Can be taken with or without food
- Pregabalin CR tablet to be administered after an evening meal
- Gradual tapering of dose when discontinuing
Interaction
- Unlikely to be involved in significant pharmacokinetic drug interactions
- No significant interactions with food
Contraindications
- Contraindicated in patients with known hypersensitivity to Pregabalin

- No adequate and well-controlled studies in pregnant women

- Caution with concomitant CNS depressants
