Indications
- Short-term management of post-operative pain
- Relieves pain, inflammation, and light sensitivity after ocular surgery
- May be used to decrease eye inflammation or redness caused by seasonal allergies
Description
- Belongs to the pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
- Chemical composition: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
- Inhibits synthesis of prostaglandins
- Biological activity associated with the S-form
- Linear pharmacokinetic property
- Highly protein bound
- Mainly metabolized in the liver
- Metabolites and some unchanged drugs excreted in the urine
Pharmacology
- Potent analgesic
- Inhibits cyclooxygenase enzyme system
- Minimal anti-inflammatory effect at analgesic dose
Dosage & Administration
- Tablet: Recommended dose is 10 mg every 4-6 hours for short-term use (up to 7 days)
- Injection: Administer as single or multiple doses for moderately severe, acute pain
- Onset of analgesic effect: Within 30 minutes with maximum effect in 1 to 2 hours after dosing IV or IM
- Duration of analgesic effect: 4 to 6 hours
Interaction
- Interaction with other NSAIDs or aspirin: Increase side effects
- Interaction with anti-coagulants: Enhance anti-coagulant effect
- Interaction with Beta Blocker: Reduce anti-hypertensive effect
- Interaction with ACE Inhibitors: Increase risk of renal impairment
- Interaction with Methotrexate: Enhance toxicity of methotrexate
Contraindications
- Hypersensitivity to Ketorolac or other NSAIDs
- Children under 16 years of age
- Prophylactic analgesic before surgery

Precautions & Warnings
- Exercise caution in patients over 65 years
- Caution in patients with active or suspected peptic ulcer or gastrointestinal bleeding
- Caution in patients with asthma and liver dysfunction
- For eye drops: Discontinue use if evidence of corneal epithelial breakdown, avoid use with contact lenses
