Indications

  • Reduces blood sugar levels
  • Used in type 2 diabetes mellitus in adults
  • When diet, exercise, and weight loss alone are not sufficient

Pharmacology

  • Stimulates insulin release from pancreatic cells
  • Facilitates Ca2+ transport across beta-cell membranes
  • Decreases hepatic glucose output

Dosage & Administration

  • Usual initial dose is 40 to 80 mg daily
  • Dose can be increased up to 320 mg daily
  • Recommended daily dose is one to four tablets (maximum 120 mg) in a single intake at breakfast time
  • Should be taken before meal
  • Not used in children

Interaction

  • May interact with oral antidiabetics, GLP-1 receptor agonists, insulin, antibiotics, beta-blockers, ACE-inhibitors, antifungal medicines, antidepressants, painkillers, alcohol, etc.

Contraindications

  • Allergic to gliclazide or related medicines
  • Insulin-dependent diabetes (type 1)
  • Diabetic ketoacidosis
  • Severe kidney or liver disease
  • Breastfeeding
