Description
- A medicine used for the treatment of erectile dysfunction and pulmonary arterial hypertension
Dosage instructions
- Recommended dose for erectile dysfunction: 50 mg taken 1 hour before sexual activity, with the possibility of adjustment to a maximum of 100 mg or a minimum of 25 mg. Maximum dosing frequency is once per day.

Pharmacological action
- Selective, reversible inhibitor of cyclic guanosine monophosphate (cGMP)-specific phosphodiesterase type 5 (PDE5)
Mode of action
- Facilitates smooth muscle relaxation and increased blood flow to the penile tissues when activated by nitric oxide released during sexual stimulation
Contraindications
- Hypersensitivity to any component of the medication
- Concomitant use with organic nitrates
Interactions
- Metabolism is mediated by cytochrome P450 isoforms 3A4 and 2C9. Inhibitors of these isoenzymes can increase Yogra clearance, while inducers may decrease Yogra clearance
Precautions warnings
- Caution when co-administered with alpha-blockers due to potential additive effect on blood pressure
- Consideration should be given to patients with underlying cardiovascular conditions, bleeding disorders, or active peptic ulceration
Pregnancy lactation
- Pregnancy category B; not indicated for use by women
- No evidence of teratogenicity or embryotoxicity in animal studies
Side effects
- Various side effects across different body systems, including cardiovascular, digestive, nervous, respiratory, skin, and urogenital systems
Overdose effects
- Adverse events at higher incidence rates and severity with doses exceeding 800 mg

Plasma levels
- Factors associated with increased plasma levels include age >65, hepatic impairment, severe renal impairment, and concomitant use of potent cytochrome P450 3A4 inhibitors
Absorption variability
- Rate of absorption reduced when taken with a high fat meal
Pharmacokinetics interaction
- Strong CYP3A4 inhibitors are expected to have greater effects on Yogra clearance

Potential complications

Adverse effects high doses
- Visual disturbances, decreased blood pressure, syncope, and prolonged erection at high doses
Overdose treatment
