Description

  • A medicine used for the treatment of erectile dysfunction and pulmonary arterial hypertension

Dosage instructions

  • Recommended dose for erectile dysfunction: 50 mg taken 1 hour before sexual activity, with the possibility of adjustment to a maximum of 100 mg or a minimum of 25 mg. Maximum dosing frequency is once per day.

Pharmacological action

  • Selective, reversible inhibitor of cyclic guanosine monophosphate (cGMP)-specific phosphodiesterase type 5 (PDE5)

Mode of action

  • Facilitates smooth muscle relaxation and increased blood flow to the penile tissues when activated by nitric oxide released during sexual stimulation

Contraindications

  • Hypersensitivity to any component of the medication
  • Concomitant use with organic nitrates

Interactions

  • Metabolism is mediated by cytochrome P450 isoforms 3A4 and 2C9. Inhibitors of these isoenzymes can increase Yogra clearance, while inducers may decrease Yogra clearance

Precautions warnings

  • Caution when co-administered with alpha-blockers due to potential additive effect on blood pressure
  • Consideration should be given to patients with underlying cardiovascular conditions, bleeding disorders, or active peptic ulceration

Pregnancy lactation

  • Pregnancy category B; not indicated for use by women
  • No evidence of teratogenicity or embryotoxicity in animal studies

Overdose effects
