Indications
- Bronchial asthma
- Bronchospasm
- Chronic obstructive pulmonary disease (COPD)
- Pulmonary disease with spastic bronchial component
Pharmacology
- Novel bronchodilator
- Inhibits phosphodiesterase 4
- Relaxes bronchial smooth muscle
- Decreased affinities toward adenosine A1 and A2 receptors
- Inhibits platelet activating factor (PAF)
- Inhibits leukotriene production
Dosage & Administration
- Dosage for Elderly: 200 mg tablet two or three times daily
- Dosage for Adults: 400 mg tablet two or three times daily
- Dosage for Children >12 years of age: 10 ml syrup or 200 mg tablet two or three times daily
- Dosage for Children 6-12 years of age: 6-9 mg/kg body weight two times daily
- Doxophylline SR tablet to be taken once daily if required daily dose is 400 mg
Interaction
- Not to be administered with other xanthine preparations
- Limit consumption of beverages and food containing caffeine
- Caution with ephedrine or other sympathomimetic drugs
- Concurrent administration of erythromycin, TAO, lincomycin, clindamycin, allopurinol, cimetidine, influenza vaccine and propanolol may reduce hepatic clearance of xanthine derivatives
- Phenytoin, other anticonvulsants and cigarette smoking may increase the clearance of xanthine derivatives
Contraindications
- Known hypersensitivity to the drug or other xanthine derivatives
- Acute myocardial infarction
- Hypotension
- During lactation

Pregnancy & Lactation
- Animal tests have shown no interference with pre and postnatal growth
- Use during pregnancy should be carefully evaluated case by case
- Contraindicated during lactation
- No effects on ability to drive and use machines
Precautions & Warnings
- Factors may reduce hepatic clearance of xanthine derivatives
- Monitoring of blood levels recommended in certain cases
- Caution in patients with cardiac disease, hypertension, elderly, severe hypoxemia, hyperthyroidism, chronic corpulmonale, congestive heart failure, liver disease, peptic ulcer, and renal impairment
- No risk of addiction or dependence
Overdose Effects
- Severe cardiac arrhythmias
- Tonic-clonic seizure
- Discontinuation of treatment may be required
- No specific antidote
- Symptomatic treatment of cardiovascular collapse
