Composition

  • 375 mg tablet: Amoxicillin 250 mg as Amoxicillin Trihydrate BP, Clavulanic Acid 125 mg as Diluted Potassium Clavulanate BP
  • 625 mg tablet: Amoxicillin 500 mg as Amoxicillin Trihydrate BP, Clavulanic Acid 125 mg as Diluted Potassium Clavulanate BP
  • 1 gm tablet: Amoxicillin 875 mg as Amoxicillin Trihydrate BP, Clavulanic Acid 125 mg as Diluted Potassium Clavulanate BP
  • Powder for Suspension: Amoxicillin 125 mg as Amoxicillin Trihydrate BP, Clavulanic Acid 31.25 mg as Diluted Potassium Clavulanate BP
  • Powder for Suspension (Forte): Amoxicillin 400 mg as Amoxicillin Trihydrate BP, Clavulanic Acid 57.5 mg as Diluted Potassium Clavulanate BP
  • 1.2 gm Injection: Amoxicillin Sodium BP (equivalent to Amoxicillin 1 gm), Clavulanate Potassium USP (equivalent to Clavulanic Acid 200 mg)
  • 0.6 gm Injection: Amoxicillin Sodium BP (equivalent to Amoxicillin 500 mg), Clavulanate Potassium USP (equivalent to Clavulanic Acid 100 mg)

Pharmacology

  • Pharmacodynamic properties: Antibacterial combination of Amoxicillin and Clavulanic Acid
  • Pharmacokinetic properties: Closely matched pharmacokinetics of both components, peak serum levels after oral administration, optimized absorption at the start of a meal, low serum binding

Dosage

  • Adults and children over 12 years:
    • Tablet: Usual adult dose of 625 mg Tablet every 12 hours or 375 mg Tablet every 8 hours
    • Suspension:
      • Children 6-12 years: 2 teaspoonful every 8 hours
      • Children 1-6years: 1 teaspoonful every 8 hours
      • Children below 1 year: 25 mg/kg/day in divided doses every 8 hours
  • Children of 2 to 12 years:
    • Mild to moderate infections:
      • 25/3.6 mg/kg/day (Suspension)
      • 2-6 years: 2.5 ml suspension b.i.d
      • 7-12 years: 5 ml suspension b.i.d
    • Serious infections:
      • 45/6.4 mg/kg/day (Forte Suspension)
      • 2-6 years: 5 ml suspension b.i.d
      • 7-12 years: 10 ml suspension b.i.d

Administration

Oral dosage form can be taken without regard to meals, IV injection suitable for intravenous administration

Interaction

  • Prolongation of bleeding time and prothrombin time
  • Reduction in efficacy of oral contraceptives
  • Concomitant use of allopurinol can increase allergic skin reactions

Contraindications

History of Penicillin hypersensitivity, cross-sensitivity with other beta-lactam antibiotics, history of Co-amoxiclav or Penicillin-associated cholestatic jaundice

Side Effects

  • Diarrhoea, pseudomembranous colitis, indigestion, nausea, vomiting
  • Hepatitis, cholestatic jaundice
  • Urticarial and erythematous rashes
  • Angioedema and anaphylaxis

Pregnancy & Lactation

Not recommended in pregnancy unless considered essential, trace quantities in breast milk

Precautions & Warnings

Use with care in patients on anticoagulation therapy or with severe hepatic dysfunction, dosage adjustment in case of moderate or severe renal impairment

Use in Special Populations

Dose adjustment for renal impairment in adults and children, caution in hepatic impairment

Overdose Effects

Unlikely to occur, gastrointestinal symptoms and electrolyte imbalance possible if overdosed

IV injection reconstituted with Water for Injection BP, not suitable for reconstitution or mixing with specific solutions

Storage Conditions

Storage below 25°C, refrigerator storage for reconstituted suspension, discard unused reconstituted vial after 20 minutes
