Indications

• Ringworm of the scalp
• Ringworm of the body
• Athlete's foot
• Ringworm of the nails
• Ringworm of the thigh
• Barber's itch
• Inhibits growth of fungi
• Identifies responsible fungi

Pharmacology

• Fungistatic
• Inhibits fungal cell mitosis
• Inhibits nuclear acid synthesis
• Binds to spindle and cytoplasmic microtubules
• Binds to alpha and beta tubulin
• Binds to human keratin and fungal microtubules

Dosage

• Accurate diagnosis of infecting organism essential
• Medication continuation till eradication
• Different treatment periods for various conditions
• Hygiene measures for infection control
• Adult dosage: 500mg daily for most infections
• Children dosage based on body weight

Administration

• Should be taken with food
• Take immediately after meals

Interaction

• May require warfarin dosage adjustment
• Concomitant use of barbiturates depresses activity
• Reduces efficacy of oral contraceptives
• Increases incidence of breakthrough bleeding

Contraindications

• Contraindicated in porphyria, hepatocellular failure
• Contraindicated in history of hypersensitivity to griseofulvin
• Conjoined twins reported in pregnancy
• Not prescribed in pregnancy

Side Effects

• Hypersensitivity reactions such as skin rashes
• Rarely angioneurotic edema or erythema
• Paresthesias of hands and feet
• Occasional oral thrush, nausea, vomiting
• Rarely proteinuria and leukopenia
• Withdrawal if granulocytopenia occurs

Pregnancy & Lactation

• Pregnancy Category C
• Adverse effects in animal studies
• Drugs given if benefit justifies risk to fetus

Precautions & Warnings

• Close observation during prolonged therapy
• Periodic monitoring of organ system function
• Possibility of cross sensitivity with penicillin
• Photosensitivity reaction warning
