Interaction
- Concomitant administration of anticholinergic drugs may antagonize the antidyspeptic effect
- Antacids and antisecretory drugs should not be given simultaneously
- Main metabolic pathway is through CYP3A4
- Concomitant use of drugs that significantly inhibit CYP3A4 may result in increased plasma levels of domperidone
Contraindications
- Known hypersensitivity to the drug
- Neonates
- Gastrointestinal hemorrhage, mechanical obstruction, perforation
- Prolactin releasing pituitary tumor (prolactinoma)
Side Effects
- Transient intestinal cramps
- Rare extrapyramidal phenomena
- Increase in plasma prolactin level
- Possible occurrence of neurological side effects in infants
- Rare allergic reactions such as rash and urticaria
Pregnancy & Lactation
- Should only be used during the first trimester of pregnancy if justified by therapeutic benefit
- Excreted in breast milk of lactating rats
- Domperidone concentrations in breast milk are 4 times lower than corresponding plasma concentrations
Precautions & Warnings
- Use with caution in children due to increased risk of extra-pyramidal reactions
- Use with caution in patients with hepatic impairment
