Indications

Pharmacology

  • Proton pump inhibitor
  • Suppression of gastric acid secretion
  • Specific inhibition of the H+/K+-ATPase
  • First single optical isomer of proton pump inhibitor
  • Better acid control than racemic proton pump inhibitors
  • Enteric-coated pellet formulation of esomeprazole magnesium
  • Metabolized in the liver by the cytochrome P450 (CYP) enzyme system
  • Plasma protein binding of 97%
  • Absorbed through oral administration
  • Excretion in the urine

Dosage

  • Oral dosage form
  • Healing of Erosive Esophagitis: 20 mg or 40 mg Once Daily for 4-8 Weeks
  • Maintenance of Healing of Erosive Esophagitis: 20 mg Once Daily
  • Symptomatic GERD: 20 mg Once Daily for 4 Weeks
  • Helicobacter Pylori eradication: Triple Therapy
  • IV Injection or IV infusion for certain patients

Administration

  • Swallowed whole and taken one hour before a meal
  • Administration of oral suspension
  • IV Injection: Intravenously over a period of at least 3 minutes

Interaction

  • Extensively metabolized in the liver by CYP2C19 and CYP3A4
  • No clinically relevant interactions with specific drugs

Contraindications

  • Known hypersensitivity to any of the formulation

Pregnancy & Lactation

Use in Special Populations

  • Safety and effectiveness in paediatric patients have not been established
  • No overall differences in safety and efficacy in the elderly
  • No dosage adjustment for mild to moderate hepatic insufficiency

Overdose Effects

  • Acute toxicity in rats
  • No reports of overdose with Creso
  • No specific antidote known
