Indications

Composition
- 133.333 mg (15%w/w) of Esomeprazole enteric coated pellets equivalent to 20 mg Esomeprazole (as Esomeprazole Magnesium Trihydrate USP)
- 266.67 mg (15% w/w) of Esomeprazole enteric coated pellets equivalent to 40 mg Esomeprazole (as Esomeprazole Magnesium Trihydrate USP)

Pharmacology
Inhibits gastric acid by blocking the hydrogen-potassium adenosine triphosphatase enzyme system
Dosage
- Erosive esophagitis: 40 mg once daily for adults (≥18 years), 20 mg once daily for children & adolescents (12-18 years)
- Maintenance of healing of erosive esophagitis: 20 mg once daily for adults (≥18 years), 20 mg once daily for children & adolescents (12-18 years)
- Risk reduction in NSAID associated gastric ulcer: 20 mg once daily for adults (≥18 years) for 4-8 weeks
- H. pylori eradication: 20 mg twice daily for adults (≥18 years), 20 mg twice daily for children & adolescents (12-18 years) for 7 days
- Zollinger-Ellison syndrome and idiopathic hypersecretion: 40-80 mg twice daily for adults (≥18 years)
- Children 1-11 years: 10 mg once daily for 8 weeks (weight <20 kg), 10 mg or 20 mg once daily for 8 weeks (weight ≥20 kg)
Administration
Tablets should be swallowed whole with liquid, and should not be chewed or crushed. Can also be dispersed in half a glass of non-carbonated water, stirred until the tablets disintegrate and drunk immediately or within 30 minutes.
Interaction
Extensively metabolized in the liver, not likely to inhibit certain CYP enzymes. Potential interference with CYP2C19, may interfere with the absorption of drugs where gastric pH is an important determinant of bioavailability.
Contraindications
Contraindicated in patients with known hypersensitivity to any of the formulation

Pregnancy & Lactation
Caution advised, not known if Esomeprazole or its metabolites appear in human breast milk
Precautions & Warnings
- Use with caution in patients with severe liver dysfunction and severe renal impairment
- May slightly increase the risk of hip, wrist and spine fracture, particularly when taken over a period of more than one year
Storage Conditions
Store in a cool & dry place below 25°C, protect from light, keep out of reach of children
