Indications
- Severe forms of acne
- Nodular acne
- Conglobate acne
- Acne at risk of scarring

Pharmacology
- Alters progress through cell cycle
- Reduces sebum production
- Prevents blockage of pores
- Reduces growth of acne causing bacteria
- Induces apoptosis in sebocytes
Dosage & Administration
- Capsules to be taken with food
- Start dose of 0.5 mg/kg daily
- Duration of treatment: 16-24 weeks
- Therapeutic response and adverse effects are dose-related and vary between patients
- Not recommended for patients below 12 years of age
- Dose adjustment for patients with severe renal insufficiency
- Lower dose for patients with severe intolerance
Interaction
- Additive adverse effects with vitamin A or its derivatives
- Decreased efficacy with microdosed progesterone
- Increased risk of local irritation with topical keratolytic or exfoliative anti-acne agents
- Reduced efficacy with oxidising agents like benzoyl peroxide
Contraindications
- Contraindicated in pregnant or breastfeeding women
- Hypersensitivity to Isotretinoin
- Hepatic insufficiency
- Excessively elevated blood lipid values
- Hypervitaminosis A
- Concomitant treatment with tetracyclines

Pregnancy & Lactation
- Absolute contraindication to pregnancy
- Great risk of severe fetal malformations
- Increased incidence of spontaneous abortion

- Comprehensive information on pregnancy prevention for female patients
- Medically supervised pregnancy tests
- Prescribing and dispensing restrictions for women of childbearing potential
- Male patients should not share medication with females
- Patients should not donate blood during therapy and one month after treatment
- Monitoring for signs of depression and skin reactions
- Avoidance of intense sunlight and aggressive chemical dermabrasion during treatment
- Cautious use in patients with diabetes, obesity, alcoholism or lipid metabolism disorder
