Description
- This tablet combines naproxen, a non-steroidal anti-inflammatory drug (NSAID), and esomeprazole, a proton pump inhibitor, for the relief of signs and symptoms of osteoarthritis, rheumatoid arthritis, ankylosing spondylitis, and to decrease the risk of developing gastric ulcers in patients at risk of developing NSAID-associated gastric ulcers.

Dosage
- Adults: One tablet twice daily of either: 375 mg naproxen/20 mg esomeprazole; or 500 mg naproxen/20 mg esomeprazole
- Juvenile Idiopathic Arthritis in Adolescent Patients 12 Years of Age & Older: Dosage varies based on weight
Administration
- Take tablet at least 30 minutes before meals
- Do not split, chew, crush or dissolve the tablet
Interaction
- Concomitant use of NSAIDs may reduce the antihypertensive effect of certain medications
- Concomitant use with warfarin may result in increased risk of bleeding
- May interfere with the absorption of drugs where gastric pH is an important determinant of bioavailability
- Administration with high-fat food may affect absorption
Contraindications
- Known hypersensitivity to any component of the tablet
- History of asthma, urticaria, or other allergic-type reactions after taking aspirin or other NSAIDs
- Use during the peri-operative period in the setting of coronary artery bypass graft (CABG) surgery
Side Effects
- Gastrointestinal side effects reduced compared to naproxen alone
- Possible increased risk of arterial thrombotic events
- Reported adverse events include nausea, vomiting, diarrhea, constipation, abdominal pain, GI bleeding, and gastritis
Pregnancy & Lactation
- Not recommended during the third trimester of pregnancy
- Should not be used during breastfeeding
- Use may impair female fertility
Precautions & Warnings
- Avoid use with other NSAIDs, especially cyclooxygenase-2 selective inhibitors
- Minimize dose and duration of treatment to reduce adverse effects
- Monitor high-risk patients closely for gastrointestinal complications, cardiovascular effects, and renal effects
Use in Special Populations
- Use caution and possible dosage adjustment in elderly patients
- Not recommended for patients with moderate to severe or severe renal impairment
- Monitor patients with mild to moderate hepatic impairment closely
Overdose Effects
- Symptoms may include lethargy, dizziness, gastrointestinal discomfort, renal dysfunction, respiratory depression
- No specific antidotes
- Manage with symptomatic and supportive care
