Description
- Baclofen is indicated for spasticity resulting from multiple sclerosis, flexor spasms, concomitant pain, clonus, muscular rigidity, skeletal muscle spasm resulting from rheumatic disorders, spinal cord injuries, spinal cord diseases, cerebrovascular accidents, neoplastic brain diseases.
- Baclofen inhibits both monosynaptic and polysynaptic reflexes at the spinal level by stimulating the GABA₋ receptors, producing CNS depression, and exerting an antinociceptive effect.
- Dosage & Administration: Adult & children over 10 years- 5 mg 3 times daily, max.100 mg daily. Children: treatment & maintenance doses vary with age
- Interaction: Baclofen may interact with agents acting on the central nervous system, alcohol, synthetic opiates, antihypertensives, tricyclic antidepressants, levodopa, carbidopa, MAO inhibitors, magnesium sulfate, and neuromuscular blocking agents.
- Contraindications: Hypersensitivity to any component of Baclof.
- Side Effects: Transient drowsiness, daytime sedation, dizziness, weakness, fatigue, headache, insomnia, euphoria, depression, hypotension, nausea, constipation, urinary frequency, rash, pruritus, weight gain, hepatic function disorders.
- Pregnancy & Lactation: Category B3, safe use not established, excreted in breast milk.
- Precautions & Warnings: Lower doses for impaired renal function, caution in patients with psychotic disorders, schizophrenia, depressive or manic disorders, epilepsy, muscle spasticity, peptic ulcers, cerebrovascular diseases, hepatic, renal, or respiratory failure, cardiopulmonary disease, respiratory muscle weakness, neurogenic disturbances, preexisting sphincter hypertonia, stroke, hepatic diseases, diabetes mellitus.
- Overdose Effects: Gastric lavage is important.
- Therapeutic Class: Centrally acting Skeletal Muscle Relaxants
- Storage Conditions: Keep below 30°C, away from light & moisture, out of the reach of children.
