Description
Esprazo is a proton pump inhibitor indicated for relieving chronic heartburn symptoms, healing erosive esophagitis, maintenance of healing, H. Pylori eradication, Zollinger-Ellison Syndrome, dyspepsia, and ulcers.
Indications
- Relief from chronic heartburn symptoms
- Healing of erosive esophagitis
- Maintenance of healing of erosive esophagitis
- H. Pylori eradication
- Zollinger-Ellison Syndrome
- Acid-related dyspepsia
- Duodenal and gastric ulcer
Pharmacology
- Proton pump inhibitor suppressing gastric acid secretion
- Specific inhibition of H+/K+-ATPase
- Better acid control than racemic proton pump inhibitors
- Enteric-coated pellet formulation of esomeprazole magnesium
- Plasma protein binding
- Metabolized by CYP2C19 and CYP3A4
- Excretion in urine and feces
Dosage
- Healing of Erosive Esophagitis: 20 mg or 40 mg once daily for 4-8 weeks
- Symptomatic GERD: 20 mg once daily for 4 weeks
- H. Pylori eradication: Triple therapy with esomeprazole 40 mg once daily for 10 days
- Zollinger-Ellison Syndrome: Dose of 20-80 mg once daily
- Acid-related Dyspepsia: 20-40 mg once daily for 2-4 weeks
- Duodenal and Gastric Ulcer: 20 mg once daily for 2-8 weeks
- IV injection dosage for adult and pediatric patients
- Risk reduction of rebleeding of Gastric or Duodenal Ulcers following therapeutic endoscopy in adults
Administration
- Swallow tablet or capsule whole and take one hour before a meal
- Oral suspension preparation and administration
- IV injection preparation and administration
Interaction
- Extensive metabolism by CYP2C19 and CYP3A4
- Likely interference with absorption of drugs affected by gastric pH
- Co-administration effects with clarithromycin
- No significant interactions with certain drugs
Contraindications
Hypersensitivity to any of the formulation

- Specific instructions for patients with difficulty swallowing capsules
- Use of antacids while taking Esprazo
Use in Special Populations
- Safety and effectiveness in pediatric patients not established
- No overall differences in safety and efficacy in geriatric use
- No dosage adjustment recommended for mild to moderate hepatic insufficiency
- Pharmacokinetics not expected to be altered in renal impairment
Overdose Effects
- Acute toxicity symptoms in rats
- No reports of overdose with Esprazo
- Symptomatic and supportive treatment recommended in case of overdose
Storage Conditions
Store at a temperature not exceeding 30°C in a dry place, protect from light and moisture, keep out of reach of children
