Indications
- Prevention of nausea and vomiting associated with initial and repeat courses of emetogenic cancer chemotherapy
- Prevention and treatment of post-operative nausea and vomiting
- Prevention of radiotherapy-induced nausea and vomiting
Pharmacology
- Highly selective 5HT3 receptor-antagonist
- Mode of action in the control of nausea and vomiting not fully known
- Blocks initiation of vomiting reflex
- Antagonizes 5HT3 receptors in peripheral and central nervous system
Dosage
- Oral tablets/orodispersible tablets

- Dosage varies for different conditions and age groups
Administration
- Oral soluble film dissolves on the tongue within 20 seconds
- Does not require water for dissolution or swallowing
Interaction
- Metabolized by hepatic cytochrome P-450 drug-metabolizing enzymes
- Inhibitors or inducers of these enzymes may affect clearance and half-life of Ondansetron
Contraindications
- Hypersensitivity to the drug or its components
- Concomitant use of apomorphine
Side Effects
- Frequently reported adverse events: headache, constipation, diarrhea
- Mild or moderate majority of adverse events
- Rare cases of anaphylaxis, brochospasm, tachycardia
Pregnancy & Lactation
- No evidence of impaired fertility or harm to the fetus in rats and rabbits
- Excretion in breast milk of rats
- Caution advised when administered to nursing women
Precautions & Warnings
- Hypersensitivity reactions reported in patients allergic to other 5-HT3 receptor antagonists
- Not a stimulant for gastric or intestinal peristalsis
- Should not be used as a substitute for nasogastric suction
