Indications

Pharmacology
- Antineoplastic agent disrupting microtubular network in cells
- Promotes assembly of tubulin into stable microtubules
- Inhibits microtubule disassembly
- Binds to free tubulin, decreasing critical intracellular concentration of tubulin
- Cytotoxic against various tumor cell lines
Dosage & Administration
- Administered intravenously (IV) over 1 hr every 3 weeks
- Various dosage regimens for different indications
- Premedication with oral corticosteroids recommended
- Adjust dose as needed
Interaction
- CYP3A4 substrate
- Metabolism of docetaxel may be modified by concomitant administration of certain compounds
Contraindications
- Neutrophil counts of <1500 cells/mm3
- History of severe hypersensitivity reactions to docetaxel or polysorbate 80

Precautions & Warnings

- Higher risk of severe adverse reactions in patients with hepatic impairment
- Avoid in patients with low neutrophil counts
- Monitoring for severe hypersensitivity reactions and acute myeloid leukemia
Overdose Effects
- No known antidote for docetaxel overdose
- Complications may include bone marrow suppression, peripheral neurotoxicity, and mucositis
- Supportive measures and therapeutic G-CSF recommended in case of overdose
