Indications
- Treatment of thrombocytopenia in patients with immune thrombocytopenia
- Treatment of thrombocytopenia in patients with hepatitis C infection
- Treatment of severe aplastic anemia
Pharmacology
- Small molecule thrombopoietin (TPO) receptor agonist
- Interacts with the transmembrane domain of the human TPO-receptor
- Induces signaling cascades leading to increased platelet production
Dosage & Administration
- Route of administration: Oral intake
- Can be taken without a meal or with a meal low in calcium
- Dosage for different conditions and patient demographics
Interaction
- Take eltrombopag at least 2 hours before or 4 hours after medications or products containing polyvalent cations
- Take eltrombopag at least 2 hours before or 4 hours after calcium-rich foods
Contraindications
- Contraindicated in patients with known hypersensitivity to eltrombopag or any components
Side Effects
- Common side effects: anemia, nausea, pyrexia, ALT increased, cough, fatigue, headache, diarrhea
Pregnancy & Lactation
- Insufficient data for assessing drug-associated risks for major birth defects or adverse outcomes
- Not recommended during breastfeeding
Precautions & Warnings
- Increased risk of hepatic decompensation in chronic hepatitis C patients
- Increased risk of progression of myelodysplastic syndromes to acute myeloid leukemia
- Risk of portal vein thrombosis in patients with chronic liver disease
Use in Special Populations
- Safety and efficacy not established in pediatric patients below the age of 1 year
Overdose Effects
- Excessive increase in platelet counts may result in thrombotic complications
- Consider oral administration of metal cation preparations to limit absorption in case of overdose
