Indications
- Treatment of hypertension
- Reducing risk of stroke in patients with hypertension and left ventricular hypertrophy
Pharmacology
- Losartan and its metabolite block vasoconstrictor and aldosterone-secreting effects of angiotensin II
- Hydrochlorothiazide affects renal tubular mechanisms of electrolyte reabsorption
Dosage
- Usual starting dose of 50/12.5 is one tablet once daily
- Dosage may be increased to 100/25 once daily for non-responding patients
- Antihypertensive effect attained within three weeks after initiation of therapy
Administration
May be administered with other antihypertensive agents. Can be taken with or without food.
Interaction
- No significant drug-drug interactions found in studies
- Possible interactions with potassium-sparing diuretics, non-steroidal anti-inflammatory drugs, antidiabetic drugs, other antihypertensive drugs, cholestyramine, and colestipol resins
Contraindications
- Hypersensitivity to any component of the product
- Anuria or hypersensitivity to sulfonamide-derived drugs

- Hypokalemia may rarely develop

Use in Special Populations
- Use with caution in patients with renal or hepatic impairment
- Safety and effectiveness not established in pediatric patients
Overdose Effects
- Manifestation of overdosage could be hypotension and tachycardia
- Signs and symptoms caused by electrolyte depletion
- Supportive treatment for symptomatic hypotension
