Pharmacology
- Inhibits bacterial topoisomerase IV and DNA gyrase
- Antibacterial action
- Inhibits DNA replication
- Inhibits DNA transcription repair and recombination
- Activity against a wide range of microorganisms
Dosage
- Available in 250mg, 500mg, and 750mg tablets
- Oral administration every 24 hours
- Oral solution to be taken before or after eating
- Intravenous injection for adults and children
- Sequential therapy (intravenous to oral) may be instituted
Administration
- Instructions for use of Levofloxacin infusion
- Check for minute leaks
- Do not use if solution is cloudy or a precipitate is present
- Suspend container from hanger
- Regulate rate of administration with flow control clamp
Interaction
- Avoid co-administration with multivalent cations
- Reduction of absorption by antacids, iron, and adsorbents
- Increased risk of CNS stimulation with NSAIDs
- Increased risk of bleeding with warfarin
Contraindications
- History of hypersensitivity to levofloxacin
- Contraindicated in patients with known sensitivity to quinolone antimicrobial agents
Side Effects
- Risk of retinal detachment
- Nausea, vomiting, diarrhea, abdominal pain, flatulence
- Rare occurrence of phototoxicity (0.1%)
- Rarely seen: tremors, depression, anxiety, confusion
Pregnancy & Lactation
- Not recommended during pregnancy or nursing
- Effects on the unborn child or nursing infant are unknown
Precautions & Warnings
- Administered by slow intravenous infusion
- Adequate amount of water should be taken during administration
- Dose adjustment in the presence of renal insufficiency
Overdose Effects
- Low potential for acute toxicity
- Stomach should be emptied in case of acute overdosage
- Patients should be kept under observation
- Appropriate hydration should be maintained
