Indications

  • Treatment of type 2 diabetes mellitus
  • Indicated for adults and children from 10 years of age and adolescents
  • May be used as monotherapy or in combination with other oral antidiabetic agents or with insulin
  • Reduction of diabetic complications shown in overweight type 2 diabetic adult patients treated with metformin as first-line therapy after diet failure

Pharmacology

Contraindications

Precautions & Warnings

  • Substantially excreted by the kidney
  • Lowers vitamin B12 level
  • Increases risk of hypoglycemia when used in combination with insulin
  • Regular monitoring of renal function advised
  • Use not advised in conditions causing dehydration or serious infections or trauma

Use in Special Populations

  • Dosage adjustment based on renal function in elderly subjects
  • Careful follow-up of the effect of metformin on growth and puberty in metformin-treated children
  • Renal function assessment before treatment initiation and regularly thereafter

Overdose Effects

  • Hypoglycemia not seen with Diafit doses up to 85 gm
  • Lactic acidosis occurred in such circumstances
  • Lactic acidosis is a medical emergency and must be treated in hospital
  • Hemodialysis is the most effective method to remove lactate and Diafit
