Indications
- Treatment of chronic HCV genotype 1, 2, 3, 4, 5, or 6 infection
- Treatment for adult and pediatric patients 3 years of age and older
- Treatment with or without cirrhosis
- Treatment with compensated cirrhosis
- Treatment with decompensated cirrhosis in combination with ribavirin
Pharmacology
- Inhibitor of HCV NS5B RNA-dependent RNA polymerase
- Inhibitor of HCV NS5A protein
- Mechanism of action involving chain termination of HCV RNA
- No clinically relevant effect on QTc interval at recommended dosage
Dosage & Administration
- Recommended treatment regimen and duration based on patient population
- Recommended dosage for adults and pediatric patients
- Ribavirin dosage modifications based on hemoglobin and creatinine clearance
Interaction
- Potential for drug interactions with P-gp and BCRP substrates
- Potential for drug interactions with CYP inducers and inhibitors
- Established or potentially significant drug interactions
Contraindications
- Contraindicated in patients for whom Ribavirin is contraindicated
Side Effects
- Common side effects: fatigue, nausea, headache, anemia, diarrhea, insomnia
- Rare adverse events including reduced hemoglobin level and reduced platelet count
- Serious symptomatic bradycardia reported when coadministered with Amiodarone
Pregnancy & Lactation
- Contraindicated in pregnant women when administered with Ribavirin
- Risk of effects on the breastfed infant
Precautions & Warnings
- Risk of Hepatitis B Virus Reactivation in coinfected patients
- Risk of serious symptomatic bradycardia when coadministered with Amiodarone
