Indications

• Treatment of chronic HCV genotype infection
• Treatment without cirrhosis
• Treatment with compensated cirrhosis
• Prior treatment with NS5A inhibitor or NS3/4A protease inhibitor

Pharmacology

• Inhibition of HCV NS3/4A protease
• Inhibition of HCV NS5A protein
• Disruption of viral life cycle
• Support of HCV replicase complex

Dosage & Administration

• Recommended daily dosage: glecaprevir 300mg and pibrentasvir 120mg
• Recommended treatment duration based on patient type and genotype

Interaction

• Potential interactions with carbamazepine, efavirenz, and St. John’s wort

Contraindications

• Severe hepatic impairment (Child-Pugh C)
• Coadministration with atazanavir and rifampin

Side Effects

• Common adverse reactions: headache, fatigue

Pregnancy & Lactation

• No adequate human data available for pregnancy risk assessment
• Unknown excretion of components in human breast milk
• Consideration of breastfeeding benefits and maternal clinical need

Precautions & Warnings

• Risk of Hepatitis B Virus Reactivation
• Monitoring for HBV reactivation and hepatitis flare in coinfected patients

Use in Special Populations

• No dosage adjustment required for renal impairment
• Dosage contraindicated for moderate hepatic impairment

Overdose Effects

• Monitoring for signs and symptoms of toxicities
• Symptomatic treatment in case of overdose
