Indications
- Treatment of moderate to severe painful conditions
- Postoperative pain
- Colic and spastic pain
- Cancer pain
- Joint pain
- Neck and back pain
- Pain associated with osteoporosis
Pharmacology
- Centrally acting synthetic analgesic compound
- Inhibits reuptake of neurotransmitters
- Modifies transmission of pain impulses
- Stimulation of mu-opioid receptors
- Modulation of central monoaminergic inhibitory pathways
Dosage & Administration
- Usual doses of 50 to 100 mg every four to six hours
- Initial dose of 100 mg for acute pain
- Initial dose of 50 mg for chronic painful conditions
- Total daily dosage by mouth should not exceed 400 mg
- One sustained release capsule or tablet every 12 hours
- Injection dose of 50-100 mg every 4 to 6 hours
- Suppository dose of 100 mg Tramadol Hydrochloride 6 hourly
- Higher daily doses for cancer pain and severe pain after operations
Interaction
- Monoamine oxidase (MAO) inhibitors not recommended for combination
- Concomitant administration of carbamazepine causes significant increase in Dolorex metabolism
Contraindications
- Hypersensitivity to the drug
- Contraindicated in acute intoxication with certain substances

Pregnancy & Lactation

Precautions & Warnings
- Respiratory depression risk
- Not recommended for patients dependent on opioids
- Caution with concomitant CNS depressants
- Caution with concomitant MAO inhibitors
- Use caution in patients with increased intracranial pressure, head injury, acute abdominal conditions
Use in Special Populations
- In children from the age of 1 year Dolorex can be given in a dose of 1-2 mg/kg body weight
- Suppository (100 mg Dolorex) not for children below the age of 14 years
- Dolorex 100 mg SR Capsules not studied in children
