Indications
- Short-term management of moderate to severe acute post-operative pain
- Relieves pain, inflammation, and light sensitivity after ocular surgery
- Decrease eye inflammation or redness caused by seasonal allergies
Description
- Belongs to pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
- Chemical name: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol
- Inhibits synthesis of prostaglandins
- Biological activity associated with the S-form
- Linear pharmacokinetic property
- Highly protein bound
- Mainly metabolized in the liver
- Metabolites and some unchanged drugs excreted in the urine
Pharmacology
- Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
- Inhibits the cyclooxygenase enzyme system
- Minimal anti-inflammatory effect at analgesic dose
Dosage & Administration
- Tablet: Recommended dose is 10 mg every 4-6 hours for short-term use, not recommended for chronic use
- Injection: Use for moderately severe, acute pain, postoperative setting, with specific doses for different age groups and conditions
- Eye Drops: Adults should use 1 drop in each eye 4 times daily
Interaction
- Other NSAIDs or aspirin increase the side effects of Perilac
- Anti-coagulants enhance anti-coagulant effect
- Beta Blocker reduces the anti-hypertensive effect
- ACE Inhibitors increase the risk of renal impairment
- Methotrexate enhances the toxicity of methotrexate
Contraindications
- Hypersensitivity to Ketorolac or other NSAIDs
- Not for use in children under 16 years of age
- Should not be used as prophylactic analgesic before surgery

Precautions & Warnings
- Caution in patients over 65 years
- Caution in patients with active or suspected peptic ulcer or gastrointestinal bleeding
- Caution in patients with asthma and liver dysfunction
- Discontinue use of topical NSAIDs if evidence of corneal epithelial breakdown is present and closely monitor corneal health
- Avoid administering eye drops while wearing contact lenses
