Pharmacology
- Semisynthetic tetracycline antibiotic
- Broad spectrum activity
- Active against Staphylococcus aureus and Nocardia
- Action on protein synthesis
- Active against Haemophilus influenzae
- Active against E. coli, Proteus mirabilis, and Klebsiella
Dosage
- Usual dose: 200 mg on first day, then 100 mg daily for 7-10 days
- Severe infections: 200 mg daily for 10 days
- Acne: 100 mg daily
- Uncomplicated genital chlamydia: 100 mg twice daily for 7-21 days
Administration
- Capsules should be swallowed whole with plenty of fluid during meals while sitting or standing
Interactions
- Impaired absorption with antacids containing aluminium, calcium or magnesium, and iron-containing preparations
- Impaired absorption with bismuth salicylate
- Decreased half-life with barbiturates, carbamazepine and phenytoin
- Render oral contraceptive less effective when used concurrently with tetracyclines
- Anticoagulant therapy may require downward adjustment of dosages
- Avoid giving tetracyclines in conjunction with penicillin
Contraindications
- Hypersensitivity to tetracyclines
- Children under 8 years of age
- Pregnant women
- Lactating mothers

- Hemolytic anaemia
- Eosinophilia
Pregnancy and lactation
- Avoided in pregnant women due to risk of staining and effect on bone growth in the fetus
- Enter breast milk, hence not recommended for breastfeeding mothers

- Avoid use during tooth development to prevent permanent tooth discoloration
