- First generation Cephalosporins
Indications
- Treatment of infections caused by sensitive Gram-positive and Gram-negative bacteria
- Upper respiratory tract infections such as sinusitis, pharyngitis, tonsillitis, laryngo-tracheo bronchitis, and otitis media
- Lower respiratory tract infections including bronchitis, lobar pneumonia, and bronchopneumonia
- Urinary tract infections such as cystitis, urethritis, and pyelonephritis
- Skin and soft tissue infections like abscess, cellulitis, furunculosis, and impetigo
Pharmacology
- Semisynthetic broad spectrum bactericidal antibiotic
- Active against both gram-positive and gram-negative microorganisms
- Inhibits cross-linking process in the cell wall of bacteria
Dosage & Administration
- For oral administration in adults and children
- For injectable administration in adults and children
- Dosage adjustment necessary in renal impairment
Interaction
- Concomitant use of nephrotoxic drugs may increase the risk of kidney damage
- Diuretics and probenecid enhanced possibility of renal toxicity
Contraindications
- Not to be used in patients with known or suspected hypersensitivity to cephalosporins
Side Effects
- Gastro-intestinal disturbances
- Hypersensitivity phenomena
- Rare side effects like glossitis, heartburn, dizziness, nausea, vomiting, skin reactions, joint pains, and oedema
Pregnancy & Lactation
- Safety in pregnancy not established
- Excreted in breast milk
- Caution advised in lactating mothers
Precautions & Warnings
- Prolonged use may result in the development of superinfection
- Administered with care to patients hypersensitive to penicillins
- May cause a false positive urine glucose result
Use in Special Populations
- Recommended doses for patients with renal impairment
- Regular monitoring required for chronic urinary tract infections
- Paediatric dosages should not exceed those specified for adults
Overdose Effects
- Non-specific symptoms such as nausea, vomiting, and diarrhoea
- Supportive treatment recommended
Storage Conditions
- Suspension should be freshly prepared
- Reconstituted Suspension should be used within 7-14 days
- Injection solutions should be used within 2 hours when kept at room temperature
- Solutions retain potency for 12 hours when stored at 5°C
- Do not use later than the date of expiry
